uhp-directed, glucose 6-phosphate membrane receptor in Escherichia coli.

چکیده
Membrane vesicles were characterized for their ability to specifically bind [14C]glucose 6-phosphate. Membranes prepared from a strain carrying a ColE1 uhp hybrid plasmid showed significantly enhanced glucose 6-phosphate binding. It is hypothesized that glucose 6-phosphate binding to these membranes is due to a uhpR-directed, membrane-bound receptor which functions in regulation of the inducible uhpT gene product: the hexose phosphate permease.

Role of the phosphotransferase system in Escherichia coli strains deficient in hexose phosphate transport.
In Escherichia coli, glucose 6-phosphate is transported via the Uhp system which is inducible by glucose 6-phosphate. We showed that, in a uhp-deficient strain, glucose 6-phosphate was dephosphorylated in the periplasm and that the resulting glucose was subsequently transported into the cells via the phosphotransferase system. The uptake of glucose generated from glucose 6-phosphate allowed the...

The UhpABC regulatory system in enterobacteria controls the expression of the hexose phosphate transporter UhpT. Signaling is initiated through sensing of extracellular glucose 6-phosphate by membrane-bound UhpC, which in turn modulates the histidine-protein kinase UhpB. Together with the cytoplasmic response regulator UhpA, they constitute a typical two-component regulatory system based on His...
متن کاملTwo mechanisms for growth inhibition by elevated transport of sugar phosphates in Escherichia coli.
The Escherichia coli uhp T gene encodes an active transport system for sugar phosphates. When the uhp T gene was carried on a multicopy plasmid, amplified levels of transport activity occurred, and growth of these strains was inhibited upon the addition of various sugar phosphates. Two different mechanisms for this growth inhibition were distinguished. Exposure to glucose-6-phosphate, fructose-...
متن کاملGlucose-6-phosphate-dependent phosphoryl flow through the Uhp two-component regulatory system.
Expression of the UhpT sugar-phosphate transporter in Escherichia coli is regulated at the transcriptional level via the UhpABC signalling cascade. Sensing of extracellular glucose 6-phosphate (G6P), by membrane-bound UhpC, modulates a second membrane-bound protein, UhpB, resulting in autophosphorylation of a conserved histidine residue in the cytoplasmic (transmitter) domain of the latter. Sub...
متن کاملGlucose-6-Phosphate Dehydrogenase Protects Escherichia coli from Tellurite-Mediated Oxidative Stress
The tellurium oxyanion tellurite induces oxidative stress in most microorganisms. In Escherichia coli, tellurite exposure results in high levels of oxidized proteins and membrane lipid peroxides, inactivation of oxidation-sensitive enzymes and reduced glutathione content. In this work, we show that tellurite-exposed E. coli exhibits transcriptional activation of the zwf gene, encoding glucose 6...
